Subsequent testing shows that it's not specific to the first track. e.g. for LogID TJI04FRZXU , Tracks 2 and 3 out of a playlist containing 5 tracks failed to update the playcount (the failed tracks were 'One night in bangkok' and 'Ghosts'). After the sync, MMA indicated that 3 tracks updated (presumably the 3 tracks for which the count successfully updated).

Note: I tested to see whether the track in question was paired properly by changing the rating, and ratings did sync correctly.

Note: I tested this on multiple devices. The problem occurs consistently across them.

Also, when I connect the device via USB and browse the contents, I can see that the device's database shows that the tracks in question didn't play (despite the fact that they did). So it's actually a Playback issue rather than a sync issue.

Most probably related to recent changes re 0010168 ?

I can confirm this issue, I played one track (Jamie Cullum, All at Sea) fully in MMA and media.playcount has not changed in mmstore.db

Edit: the playcount increased, but one minute later. I was watching internal mmstore.db on rooted device using SQL Debugger. So the db must have been in operation memory meanwhile probably.

Played 6 tracks in MMW on Nexus 5/kitkat:
1) 01 Zum Gali Gali
2) One Night in Bangkok
3) Ghosts
4) MI-2
5) The Chicken Dan
6) Pump Up The Volume
8O0LZNH3EI - manual log after tracks finished playing

Then WiFi synced
DO2K0IUPRD - manual log after sync

--> MMW shows that only tracks 1, 5, 6 played.

The results with this sync are different than with the previous sync on this device, which means that the problem isn't specific to certain tracks. Perhaps related to whether the lock screen is active?

I've done a couple more tests and on both occasions playcount synchronization worked as expected. I have no idea as to why playcounts sometimes sync correctly, and other times don't.
